Fluorescence is a physical phenomenon in which a compound absorbs light and re-emits this as light of a usually higher wavelength. Since the wavelengths of the excitation light source and the emitted fluorescence can be separated very well, we can detect fluorescence with very high sensitivity, making it possible to visualize even single molecules. Many different fluorescent probes for cellular components have been developed, including genetically encoded probes like the Green Fluorescent Protein (GFP). For these reasons, fluorescence microscopy is a very powerful tool in Cell Biology research. See more at www.ibioseminars.org

AWESOME FLUORESCENCE THERMOCHROMISM


In this video we're going to demonstrate fluorescence thermochromism. Simply dissolve a small amount of copper iodide into pyridine to form a clear yellow solution of trispyridine copper iodide. Normally this does not fluoresce but if cooled to liquid nitrogen temperatures it will fluoresce a strong yellow color. To understand why this happens one must first understand a little about fluorescence. The ultraviolet light is absorbed by the molecule and excites electrons in it to higher energy levels. Normally the molecule bumps into other molecules and looses this energy, dropping the electrons back into their ground levels. In fluorescent materials though, the electrons release their energy first as fluorescent light before they loose their energy through collisions. In trispyridine copper iodide, the molecules vibrate and collide too much at room temperature and thus destroy any chance of fluorescence, so the substance looks non-fluorescent. But if cooled low enough, the molecules vibrate less and thus the electrons have more opportunity to emit light before a collision. Now moving on, if a few drops of the trispyrdine copper iodide solution is put on a sheet of filter paper and allowed to dry. it looses a pyridine molecule to become bispyridine copper iodide. This substance is more stable and will fluoresce at room temperature. But cooling will not improve the fluorescence noticeably since the substance is already stable enough to resist collisional deactivation. Warming <b>...</b>

Taiwan Lab Creates Fluorescent Angelfish


Glowing examples of innovative biotechnology. Fluorescent angelfish in a Taiwan laboratory. The Jy Lin Company, Taiwan's largest exporter of ornamental fish, has for the first time, successfully inserted a fluorescent protein gene into the reproductive organs of medium-sized fish, allowing them to produce radiant offspring. Company spokesman Lin Yu-Ho, explains how the process works for this particular fish family. [Lin Yu-Ho, Jy Lin Company]: "With the medium-sized cichlid fish, its biological cycle cannot be controlled by artificial lighting, plus the low number of fertilized eggs makes collection a challenge. It will take a very long time with the micro-injection method. For species like these, we deployed the method of reproductive organ electroporation. We inject fluorescent genes into the reproductive organs and leave the male and female fish to reproduce on their own, and then they can give birth to fluorescent babies." Electroporation is a process whereby an electrical pulse creates passages through cell walls for the transplantation of foreign molecules, in this case the fluorescent protein gene. The shining new breed of fish took over three years and almost 320000 US dollars to develop. And it will take some time and safety tests before discerning aquarium-fish lovers can dive in and buy their very own glow-in-the-dark angelfish.

Fluorescent DNA in Optical Tweezers


A 10 kb DNA molecule labelled with digoxigenin and biotin is tethered between Streptavidin and anti-DIG coated microspheres held in double-trap optical tweezers. The DNA is stained with SYBR-gold and observed by epi-fluorescence imaging. The video shows many experiments, some with single DNA molecules between the beads, and some with two or more. Hydroponic Medical Marijuana under fluorescent lights


I got 8 oz. of Blue Cheese medical marijuana buds from a single plant (the big one in this video) using a hydroponic system and only 50 standard 23 watt household fluorescent bulbs (1150 watts total). The buds were big and dense, and the quality was better than those produced by an expert grower (my neighbor) in dirt under a 1000 watt HPS. We both used Advanced Nutrients and the same Blue Cheese mother plant was the donor for clones.

Westcott vs ePhoto Fluorescent Softbox Showdown


www.learningdslrvideo.com In my quest for learning more about light, I want to see if there was much difference from the very expensive softbox that Westcott makes (Westcott TD6 DLX Medium Daylight Kit) coming it at a whopping $829 vs. an ePhoto $100 softbox Fluorescent light my friend let me borrow. Both images you see at the beginning are set to 5500K, ISO 200. However the Westcott was 2/3 of a stop brighter than the ePhoto. The Westcott has 6 50watt bulbs, and the ePhoto has 5 45watt bulbs. I didn't do any color correction. Only one light used at a time with no reflectors and both the same distance to the talent (me). Wow the Westcott is so expensive. I can hardly tell any difference between the two lights. Why does the Westcott cost so much more? While the ePhoto softbox works well I would not recommend it to some that travels a lot, because I don't think they are meant for the road. I think if you set them up in your studio once and never take them down then I think they are a great deal. Both of them did not have any banding issues until the shutter got up to 1/320 of a second. I didn't see any flicker at all.

HALCYONE - Fluorescence Lifetime Measurement System


Halcyone is an all-in-one-box fluorescence measurement system comprising Fluorescence Upconversion spectrometer and a Time-Correlated Single Photon Counting instrument. In the upconversion mode Halcyone has a time window of 3.3 ns and femtosecond time resolution. In the TCSPC regime the time window can be expanded up to 1 ms with 200 ps resolution.
