Tholkappiyam is the earliest extant work on Tamil grammar. It is surprising that even today, roughly 2000 years later, the language follows the rules given then. Badri Seshadri provides an overview of Phonology, Phonemics and Morphology of Tamil as detailed by Tholkappiyam's first two sections in this lecture.
